Vincristine is an anti-tumor vinca alkaloid isolated from Vinca Rosea. Vincristine is being isolated from Cantharanthus roseus plant
Chemical formula of Vinca Alkaloid : C46H56N4O10
Mechanism of action:-
- It is a Mitotic inhibitor drug
- It is a cell cycle phase specific drug
- It is active only in M-Phase
- It bind to tubuline and interfere with mitotic spindle formation and thus segregation of chromosomes in metaphase is arrested.
Pharmacokinetics:-
- It given on strict Intra venous route
- Within 15-30 minute after injection, over 90% of the drug is distributed from blood into tissue.
- The liver is the major excretory organ of vincristine in Human and Animals
- The metabolism of vinca alkaloids has been shown to be mediated by hepatic chromosome P-450 isoenzymes in the CYP 3A subfamily.
- About 80% of injected dose of vincristine sulphate excrete in faeces and 10-20% excreted in urine.
Use of Vincristine:-
It is used either alone or in combination with other anti-neoplastic drugs for treatment of following condition:-
- CTVT ( Canine transmissible venereal tumor )
- Leukaemia
- Malignant lymphoma including Hodgkin’s disease and non-Hodgkin’s lymphoma
- Multiple myeloma
- Brest carcinoma, small cell bronchogenic carcinoma
- Idiopathic thrombocytopenic ( IIP )
Dose:-
In CTVT, Vincristine sulphate given at 0.025mg/kg dose.
Vincristine sulphate given once in a week for continuous 4 week
Surface area = Body weight⁰∙⁶⁷ × K/10⁴
Here K=10 in cats while K=10.1 in dogs
Side effects:-
- Linkage into surrounding tissue during I/V administration of vincristine sulphate injection may cause considerable irritation. If extravasation occur the injection should be discontinuous immediately.
- Affect hepatic function that is why it is not given in the patient which suffer from hepatic diseases.
- Alopecia
- It affect renal function that leads to Dysuria or Urinary retention
- It affects on skin and produce anaphylactic reaction. Also produce urticarial lesion.
- Leukopenia
- Adrenal disorder
- Hypotension with dehydration
- Neurological disorder
- Oedema
- Blindness
